A simple, precise, and reliable reverse-phase high-performance liquid chromatography (RP-HPLC) method was successfully developed and optimized for the simultaneous estimation of Olanzapine and Samidorphan in bulk and pharmaceutical dosage forms. Chromatographic separation was achieved using a Platisil C18 column with a mobile phase of methanol and KH₂PO₄ buffer (70:30 v/v, pH 3.5) at a flow rate of 1.0 ml/min, with detection at 225 nm. The method provided well-resolved peaks with good resolution, acceptable symmetry, and satisfactory system suitability parameters. Validation studies performed according to ICH guidelines confirmed linearity over the concentration ranges of 20–100 µg/ml for Olanzapine and 10–50 µg/ml for Samidorphan, with correlation coefficients of 0.9998 and 0.9997, respectively. Precision studies showed %RSD values below 2, accuracy studies demonstrated recoveries within 98–102%, and sensitivity was established through low LOD and LOQ values. Robustness studies indicated that minor variations in chromatographic conditions did not significantly affect performance. Forced degradation studies under acidic, basic, oxidative, thermal, and photolytic conditions revealed degradation of both drugs, but the method effectively separated degradation products from the main peaks, confirming its stability-indicating capability. Overall, the validated RP-HPLC method is robust, reproducible, and suitable for routine quality control, assay determination, and stability studies of Olanzapine and Samidorphan in pharmaceutical formulations.

INTRODUCTION

Pharmaceutical analysis plays a pivotal role in ensuring the safety, efficacy, and quality of drug products. Stability-indicating methods (SIMs) are considered essential because they not only quantify the active pharmaceutical ingredients (APIs) but also detect and separate degradation products formed under stress conditions. A stability-indicating method is defined as a validated analytical procedure that accurately and precisely measures the active ingredients without interference from degradation products, impurities, or excipients.[1-3] The main objective of a stability indicating method is to monitor results during stability studies in order to guarantee safety, efficacy and quality. It represents also a powerful tool when investigating out-of-trend (OOT) or out-of-specification (OOS) results in quality control processes. These methods ensure that the therapeutic agent maintains its intended pharmacological activity throughout its shelf life. In modern pharmaceutical practice, combination therapies are increasingly common and offer synergistic effects, improved patient compliance, and reduced pill burden. However, the simultaneous presence of multiple APIs in a single dosage form poses analytical challenges. The concurrent analysis of two drugs requires methods that can separate, identify, and quantify each component accurately, even in the presence of degradation products. [4-5] Olanzapine molecular formula is C17H20N4S (2-Methyl-4-(4-methyl-1-piperazinyl)-10H-thieno[2,3-b] [1,5] benzodiazepine) and used as Antipsychotic used in the treatment of Schizophrenia, Bipolar disorder, Major depressive disorder by acting on Dopamine and serotonin receptors.[6-9] Samidorphan molecular formula C21H26N2O4 (5-Chloro-3-[(1R)-1-[(2S)-2-(3,3-dimethyl-1-piperidinyl) propanoyl] pyrrolidin-3-yl]-1,2-benzisoxazole) and used as Antipsychotic (Second-generation) in the treatment of Schizophrenia, bipolar disorder, major depressive disorders by acting on Dopamine D2 and serotonin 5-HT2A receptors.[10-12] The combination of olanzapine and samidorphan have several analytical challenges like difference in chemical structure, potential degradation pathways, matrix interference and sensitivity requirements. These challenges necessitate the development of a robust, validated stability-indicating method that can separate and quantify both APIs and their degradation products.[13-15]

CONCLUSION
The developed RP-HPLC method proved to be robust, reproducible, and suitable for routine quality control analysis of Olanzapine and Samidorphan in combined dosage forms. Robustness studies indicated that minor variations in chromatographic conditions did not significantly affect the results, confirming the reliability of the method. Forced degradation studies demonstrated that the method is stability-indicating, as it effectively separated degradation products under various stress conditions such as acidic, basic, oxidative, thermal, and photolytic environments. Therefore, the proposed method can be confidently applied for assay determination, stability studies, and pharmaceutical analysis of these drugs in bulk and formulation.
